Hi,
I wanted to use user login bar module,
and assign the block to secondary menu.
How do I set the location of secondary menu as region?

You could disable 'secondary links' on the theme configuration page and assign secondary links to the header region.
You'll need some CSS to set it in place of the secondary links.
