Where do I put the threecol_25_50_25_stacked layout to begin customizing it?

I just downloaded threecol_25_50_25_stacked, and I'd like to use it, but where (what folder) should I drop it in to in order to begin?

Thank in advance. Not a noob to website creation, just CMS's.

You do not say what the file is for. I have never heard of it, and you do not say where you got it. Looks like that file is a custom layout for Panels? So you need to set up Panels module and I think put in in panels/includes
Best get the hang of the basics of Drupal first, before learning Panels. You can get this layout from many themes, and you may be able to acheive the result you want without the extra complexity of Panels.
