There are not much difference except API functions and some other things.
The best way to learn Drupal 7 is start working on a project by following the drupal official documentation.

Since you are already familiar with Drupal, hence you can pick it up within very short time.

Also If you have made your own plugins for D6 you should check this http://drupal.org/node/224333 for how to convert them in to D7 modules :)
