make sure you update the site to the latest v5 (5.6 atm) then to the last v6 (rc2 atm)
and ofcourse
be sure to check the readme before upgrading
There's a few things to consider
An upgrade is possible but always make sure you have backed up your site's files AND the database.
Second: your contributed modules need to be compliant with version 6, too.
Third: Content Construction Kit, Views and Date may make it necessary to experiment a bit in the upgrading process. Most of the times you have to enable the modules first, and then the theme. Sometimes the other way around works better.
Fourth: Date module fields should always be saved individually after upgrade. So for each content type they are used in.
Fifth: Don't update too soon; sometimes you may get a bug report after a day or two.
So the best thing is to test the process on a local installation before going for the live site. WOS portable is an awesome piece of work to do the job.

Finally after much wait for
Finally after much wait for the contributed modules to be ported to 6.x, now I have upgraded my website to 6.x. See my new upgraded website at http://www.vshiksha.com
