Unfortunately, given the complexity of Panels2, this type of question is simply too broad for us to be able to address in the issue queue. I suggest playing around with the module first to get a sense of it - the UI, though complicated, does have a lot of in-line explanations. I wish I could point you to some handbooks documents that could better instruct you with this, but being that Panels2 is still in its beta phases, we haven't yet been able to pull together that documentation. The best thing I can offer you is #206792: Panels 3: Creating a new front page with Views and Panels 3.
