Original design (which is not implemented yet) is to inject the outgoing mail url with a special tag which will be interpreted when users click on the link.

If it will be possible to inject that tag into outgoing emails from simplenews then YES, counting clicks will be possible. i will consider implementing this as a more generic solution.

while it is on my to do list, i have other projects which take higher personal precedence. I am open to accepting donations or work orders in order to put this module on the top of my list.
