I had Gallery 2.3 on my Durpal 6.0, I have upgraded to Drupal 6.2 and suddenly I lost my Gallery. Do I have to reinstall G2.3?

This is the error message I get
warning: mkdir() [function.mkdir]: Permission denied in "my server path"/gallery2/modules/core/classes/GalleryPlatform.class on line 564.

Seems obvious, but did you check directory permissions of your g2data folder and configuration files? Otherwise please provide some more details. How can I reproduce the problem? When does it occur? I guess you tried to create an album or sth.?

Please be a little bit more verbose in your issue descriptions AND using "Gallery does not work" or "URL Rewrite issues" as a title is not very helpful, neither for other users with the same problem nor people trying to help you.
