After some "trial and error" with exploring many modules, I now have a boat-load of modules in my .../sites/all/modules directory. At one time or other I've activated nearly all of them. Now, I'd like to clean things up a bit and remove any modules that are not really being used by my site. Any thoughts on how to inspect my site to determine which modules are really necessary for the functionality I have?

There's nothing built in
There's nothing built in that can do that very well. Not least because there's some modules that are admin-only.
I can imagine how to add some logging function that may help a real audit, but you really need a full understanding of the Drupal module-loading process to do it meaningfully, so I dunno if it would be worthwhile.
I'd actually suggest starting again about now. You'll feel better about doing a fast rebuild from scratch on a clean site!
