I think it would be good to be able to control the display of blocks by user role.
Example: You could create admin related blocks just for the admin user role or have advertising blocks only display for anonymous user.
I would see this as tying in with the existing access control system. When I asked about this in the forums I was directed to http://drupal.org/node/3371. I really think that this functionality could be very useful if part of the drupal core.
