I would like new users to be required to enter their location information, then automatically have this available to location module. Is there a way to extract location information from a custom profile field or make location information from location module present as a mandatory item on the registration page?

In order to do this I believe you'll have to use content_profile (use the dev version to get registration form integration) to turn user profiles into standard cck nodes.
