I want to modify this theme so I can only have one right side bar, with a fixed width capable of accommodating a 300x250 advert.

Have you tried simply not assigning any blocks to the right sidebar?
The pixel width can perhaps be set in the style.css file.
Consider using the Firebug extension for Firefox to inspect CSS classes.

Beyond that, this is related to theme development.
