So far I did a bunch of test installs and played around with Drupal for a bit.

What I haven't figured out is how to set up pages like you can see on last.fm

They have a bunch of blocks which are put together differently depending on the requested page.

How do I create such pages which consist of multiple blocks as main content?

It's just a 2 col layout with lots of blocks assigned to each column. Just assign the blocks to whatever region you'd like them to appear.
