Would it be possible to place another block on the page through a setting?
It would be great to habe a block top searches or other sort of block to give the visitor more possibilities to stay on the site.

Search404 does not place any blocks, all it does is either display the left and right sidebars or it avoids the display of the sidebars altogether.

But let me see if this modification, can be done, it might take some time though.

Would be great to be able to place search404 in a panel and be able to use other regions for related information.

After discussing with some colleagues, they suggested that this functionality shouldn't be a part of the search404 module.

On a positive note, I would suggest that you use panels to achieve this.
