I'm embarking on a new project for the National Pork Board that will be known as the PIG Portal.
Required Configuration:
PHP 5.1.1
MS SQL
Drupal 4.6.5
I want to use these tools to develop clients website . Does it supported .Please suggest me a good combination.Clients are asking for php5.1 with MS SQL, is it possible ?

Drupal can work with MS
Drupal can work with MS Sql.
If you have a good DBA you wont find a problem.
There are many difference between the database queries and you should do some of it agai.
You can create the MS Sql script and add it as a new contribution here or in your web site.

I'm really sorry i was not up to date. And i read that discussion,but my view is in to whether to start the project with MS SQL by taking the risk of implementing the Abstraction layer for MS SQL and fight with the issues myself without taking Hekp of Drupal-supporters,OR strictly rejecting the System Requirements suggested by clients.So i was in view to get the as more decisions as possible , I'm sorry.

Try to convince the client to use MySQL or PostgreSQL. Since you are starting the project from scratch and there is no legacy data you will do better if you go with MySQL or PostgreSQL.
If your clients are not convinced then you will have to consider another CMS or will have to handle the MS SQL issues yourself. Why don't you conduct a test run? Give yourself 2 days to get a basic site with some modules up and running nicely with MS SQL and you will get to know if you can do the whole project with MS SQL.
A word of caution here. There are very few Drupal installations using MS SQL. If you post a question regarding MS SQL not many users may help you because (a) they might not have worked with MS SQL (b) your problem may not interest them (c) your problem may be too specific.
