I was wondering if I can build a back-end interface to assign a caption and image group gallery instead of typing rel attribute in the link...

I would like to make it easy for my customer to use it

Well if you use the image or imagefield modules, and configure lightbox2 appropriately, then it should all be automatically done for you, with no need to write another admin UI. Check out the "automatic" section of http://drupal.org/node/144488 and the docs at http://drupal.org/node/274714
