I've created my image gallery using the image module but the resulting layout is 2 by 6 (2 columns and 6 rows). Is there any way for me to change the layout to perhaps 4 by 6 instead?

It's fluid -- depends on the width of your theme or your browser.
You can change the widths set in CSS. Alternatively, you can pick a different view style.
