I would love to be able to add an option in my lightboxes where when a user clicks an image and the lightbox displays the image, below the navigation and caption it could integrate with the Tweet module (http://drupal.org/project/tweet) so users could tweet the image.

There's no plans to add this functionality at present. Perhaps you should check out the documentation on opening node content in a lightbox at http://drupal.org/node/252260
