What is the best way of letting subscribers know of new content – I would ideally like an email to be sent out when there is an update to the site that they would get it as quite a few people don’t have rss aggregators.

Aslo how do you specify these by content type so for example people only interested in "story" content can receive and email or RSS update when story content is added and users interested in blog content get updates on these?

Try the Subscriptions module... I use it on a D5 site but I see there's a release for D6.
