I just saw a website that looked very similar to twitter where users would post funny stories, and below the text were icons to Share with Facebook, Tweet! and also visitors would rank (Funny) or (Not So Funny)
It was kinda like jokes where users would give a thumbs up or down! (I am thinking they used WordPress)
I dont believe it was built with Drupal but was wondering whether this can be done and if so do you know of any similar websites to Twitter that was built with Drupal? Are there nice themes and modules for that use?

In general Twitter is a decently simple site.
Look into Facebook Style Statuses, Share To Any/Service Links, and Flag.
That should be a decent starting point.
