I upgraded to 6.x-1.0-beta4 from 6.x-1.0-beta2 (thanks for the role-based titles, by the way!), and now my custom "Override title" in my user account is no longer appearing. It still shows in the edit box in my user profile, but the "Current User Title" is blank, and it's not appearing in the forum, either.

Yep, same issue on our site as well (http://www.twcgaming.org). Role-based titles appear just fine, and the role priorities for the titles work out as well. The custom ones we set as overrides not only don't show but they hide the Role-based titles as well.
