Is it possible to use multiple layouts for different sections of a single website, that means for section1 it is two column only (content-right site bar), for section2 it is left side bar-contnet and for front page it is three column (left-content-right).

You can do this with just the blocks system in Drupal - configure the blocks to only show up in certain sections. If you use the standard 3 column layout (left - content - right) this is very easy to achieve with just the block configuration.
