On an existing media gallery, I would like to replace a asset with another asset. In order to keep the current sort order, description, tags and license settings, I don't want to remove the asset and add a new one, but I would like to simple pick a new one from the asset browser.

Have you seen http://drupal.org/project/media_update? I haven't tried it myself, but it looks as if it might do what you want.

I've tried it yes. It replaces one asset with another asset throughout all media items. I only want to change one media gallery item with another one, without replacing them in all places.

That cannot be done at the moment, and I think this is a won't fix.
