Yesterday I upgraded to rc2, after also experiencing multiple emails to our members being sent.

I created a news letter almost 24 hours ago now and as of now it hasn't been sent.

At ->admin/content/simplenews/sent
The job is showing as "Currently sending by cron"

Nothing else has been updated.
Any ideas what has happened?

I have just deleted the news item which hadnt as yet been sent (now almost 36 hrs) and compiled a new news letter (over 1/2 hour ago) which simplenews isn't sending either (after the update to RC2).

Update:
The simplenews-6.x-1.x-dev.tar.gz is also effected.

Test send of newsletter does work.

Errors reported
Newsletter not send: node does not exist (nid = ; vid = ).
0 emails sent in 0.1 seconds. (after Cron was run)

Yes, the node does exist!.

It appears that something screwed my simplenews database entry.

I manually removed all traces of the module, re-installed now ok.

Did you perhaps update while newsletters were being sent?
