What is the difference between the two?

I see the pulgins are all 6.1. Should I use activity stream 6.1 to take full advantage of them?

1.0 is the release version and you should use that.

The 2.x-dev line contains the future version of the module. It is the bleeding-edge code and the download is a daily snapshot of whatever is in version control. The code may not be stable, isn't optimized for performance, isn't as well tested, and might not even work. It should not be used in production at all.
