Trying to setup and use Genesis theme - help!

Ive downloaded and tried to follow the instructions for using Genesis as my theme. I want a layout just like as in the screenshot at the bottom of this page http://drupal.org/node/460800
After making the listed changes to the themes files, is it just a matter of reconfiguring where your blocks go?

Also, so what regions are displayed in the screenshot here http://drupal.org/node/460800 toward the bottom of the page. Is that 3 blocks being displayed in the "Header Blocks" region or the "Secondary Content" region?

No, they are Gpanels, if you read the docs on that book page it tells you how to use Gpanels to get that sort of layout (basically copy/pasting a snippet into page.tpl.php, uncomment the regions in your subthemes info file and then adding some blocks to them). Read the book page, it tells you precisely how to do it.
Header blocks region is actually in the header, and floats between the logo/site name and the search box. Secondary content is a full width region between the header and the main content columns.
